Kinetics of inhibition of butyrylcholinesterase by an aqueous extract of Cuminum cyminum

Objective: According to cholinergic hypothesis, cholinesterase inhibitors are the only drugs approved for symptomatic treatment of Alzheimer’s disease. The present study, evaluates the in-vitro butyrylcholinesterase (BChE) inhibitory activity and mode of inhibition by an aqueous extract of Cuminum cyminum using Ellman’s method. Method: Ellman’s assay was used to study the cholinesterase inhibition in-vitro. IC50 value was calculated using concentration dependent curve and Lineweaver–Burk plots were used to calculate mode of inhibition. Result: An aqueous extract of C. cyminum showed mixed competitive mode of inhibition kinetics and concentration-dependent inhibition of BChE with maximum inhibition of 66.84 ± 0.001% (SEM) at 100 µg/mL final concentration and with IC50 value of 539.69 µg/mL. Conclusion: An aqueous extract of C. cyminum seed acts as potent inhibitor of BChE, which exhibited mixed competitive inhibition kinetics and therefore can be used for potential treatment for Alzheimer’s disease in future.
